{mosimage}Target of the project is the plant of a DNA-bank in analogy to the existing gene bank of all, however predominantly rare, preservation-worthy farm animal races, which was already partly realized by the Institute for Biological Agriculture and Bio Diversity of farm animals in Thalheim.
The main objective of FADNAB (Farm Animal DNA Bank) is a comprehensive archive of isolated cryo-conserved DNA of Austrian livestock. About 100 samples per breed will be frozen to enable quick and easy analyses of gene sequences for genetic distancing of breeds, research on marker genes or identification of genes coding special properties like disease resistance, fertility or special product traits.
Links between the DNA-Bank and the Austrian Gene Bank for Endangered Farm Animal Genetic Resources (semen-bank) will facilitate genetic characterisation and enable more sustainable conservation breeding plans for endangered breeds.
Collection of samples containing DNA is rather simple. The best results are obtained with blood or tissue samples like ear clippings. In birds a few quills suffice.
Existing methods for isolating and conserving DNA vary according to the analytic goals. Usually, DNA-fragments up to 20.000 bp (base pairs) are sufficient for genetic characterisation. These fragments are obtained by cracking the nuclei with sodium-dodecylsulfate (SDS)and proteolytic splitting with k-proteinase. After phenolic extraction the DNA is precipitated by means of alcohol and can almost indefinitely be stored in liquid nitrogen.
The project will be integrated into the EFABISnet project (submitted, directive EU 870/04) in the part "CRYO".
